Vooral wanneer je het substraat koloniseert, moet je de temperatuur en de vochtigheid regelen om de groei van het mycelium te verbeteren. De sporen van paddestoelen groeien het best bij een temperatuur van 21 tot 27 graden Celsius. Sommige sporen van paddestoelen hebben zelfs een temperatuur van 30 graden Celsius nodig. Bij deze temperaturen moet je er wel voor zorgen dat het substraat vochtig genoeg is (in het beste geval moet het een temperatuur van 90% hebben).
